Effect of Barbiturates and Sedormid on the In Vitro Induction of Tryptophan pyrrolase


Abstract
The effects of eight kinds of barbiturates: and sedormid, which have the porphyria-producing property, on the in vitro induction of tryptophan pyrrohse in rat¢¥s liver homogenate which is an adaptive enzyme and has iron porphyrin prosthetic group have -been studied,
Barbital, thiopental, aprobarbital(allucate), and hexobarbital(Evipan), one of N-substituted barbiturates, were of no effect on the in nitro tryptophan. pyrrolase induction. In contrast, pentobarbital, secobarbital, amobarbital, diallyl-barbituric acid(Dial), and ~allylisopropylacetylcarbamide(sedormid) inhibited the in vitro induction by 50 to 80%. Among these inhibitory- barbiturates, pentobarbital and secobarbital, which have one metthylbutyl group in No. 5 carbon atom of cyclic ureide, were most potent in its inhibition.
From these results, it was suggested: that molecular structures responsible for the inhibition of the in vitro tryptophan pyrrolase induction might be related to acetamide derivatives.
